Microorganisms have many diverse roles in soil. This investigation compared populations, metabolic phenotypic characteristics and growth on xenobiotics by aerobic, heterotrophic bacteria from horizons within the soil profile (250 cm depth) of a Mexico silt loam (Mesic, Aeric Vertic Epiaqualf). Paired cores representing summit and toe-slope landscape positions were collected from a field (3-5% slope) in the central Missouri claypan region. The field was cropped in a rotation of corn ( Zea mays L.), wheat ( Triticum aestivum L.) and soybean ( Glycine max L. [Merr.]). Cattle were allowed to graze crop residues overwinter. Soil bacteria were enumerated from selected horizons on peptone, tryptone, glucose, and yeast extract (PTGY) medium and a 1:20 dilution of PTGY. Pure cultures of bacterial isolates were screened for metabolism of various carbon sources subjected to standard diagnostic tests. The results of enumeration and phenotypic characterizations revealed diversity in distribution and types of soil bacteria and those differences were associated with source of isolation. Bacterial counts differed among the selected horizons Ap, Btg, Btg2, 2Btg, and 3Btg in the summit landscape position. Patterns of carbohydrate utilization, presence of fluorescent pigments, aromatic ring cleavage, hydrogen sulfide transformation and pesticide metabolism yielded different bacterial phenotypes related to the original sources of isolation. Phenotypic characterization revealed that 22 and 8% of summit and toe-slope isolates, respectively, were positive for glucose oxidation while 57 and 100% of summit and toe-slope isolates, respectively, were positive for denitrification. The dominant taxa presumptively identified were Pseudomonas and Xanthomonas . Facultative anaerobes detected in summit horizons below 160 cm included genera of Erwinia , Enterobacter , and Kingella , while Enterobacter was isolated from lower toe-slope horizons C, 2C, and 3C. Enumeration and phenotypic characterization were apparently related to soil physical properties such as aeration status of the claypan soil
