Summary
Leinamycin is a novel antitumor antibiotic natural product that derives its biological activity from DNA damage. We have shown that leinamycin, in addition to its known thiol-dependent DNA-alkylation pathway, causes thiol-dependent oxidation of DNA. Hydropolysulfides, formed from reaction of leinamycin with thiols, are implicated to be the active species that convert molecular oxygen to DNA-damaging hydroxyl radicals. In addition, we show that leinamycin possesses a thiol-independent DNA-alkylation pathway; our experiments suggest that thiol-independent DNA-damage by leinamycin arises from its hydrolytic activation. Also, from leinamycin-activation by phosphines and cyanides, we provide the first convincing evidence that DNA alkylation by leinamycin proceeds through the previously postulated oxathiolanone intermediate. As an offshoot of our work on leinamycin, we have developed novel syntheses of the cleft-shaped molecule dithiosalicylide, and have applied these methodologies to the syntheses of symmetrical and unsymmetrical dithiosalicylides. These dithiosalicylides are found to form interesting arrays of self-included dimers in crystal lattice
