Summary
The existing topological and functional model for FepA was reconfigured using data generated from epitope insertion and deletion mutants. A significant alteration in this model was the rearrangement of the N-terminal 150 amino acid region that is the most strongly conserved amongst TonB dependent receptors (TonB domains). Putative cell surface and transmembrane (TM) regions within this domain were relocated to a primarily periplasmic location, where they would be more accessible for interaction with the cytoplasmic membrane protein TonB. Another significant alteration was a change in the number of TM strands that constitute the outer membrane FepA Ý-barrel from 29 to just 22. In addition, several cell surface exposed regions that are potentially involved in ligand binding, either directly, or indirectly via a conformational change, were identified. Most of the epitope insertions in the N-terminal TonB conserved region, eliminated the functional capability of the protein. They also affected the surface conformation of the receptor, resulting in reduced ligand binding. Combined structural and functional analyses of fepA deletion mutants indicated the possibility of a 'dual' character for FepA, wherein the N-terminal conserved, periplasmic region is critical for function, while the C-terminal region determines FepA folding. These studies indicate that the native structure of FepA is determined by an intramolecular interaction between these two regions
