There is considerable interest in psychiatry in using neurobiological information to understand neuropsychiatric disorders, by relating genome/proteome information to the behavioral level. A framework is developed for modeling the reward mechanism of the brain using knowledge about the neurobiological mechanisms both at the cellular and the genome/proteome levels. The proposed framework consisted of a typical nucleus accumbens (NAcc) neuron with dendrites which carried 'input'' information in to the soma from other neurons, with the 'output' of the neuron being carried away by the axon. The neuron model has potassium, calcium and sodium channels and exhibits bistability. A methodology is proposed to model the behavioral aspects associated with the reward pathway and the learning of the cue-seeking behavior for a simplified example case of cocaine addiction. Results using the example case show the feasibility of the proposed approach. The entire computational framework is developed using the GENESIS software package. An approach to model the intracellular mechanisms in an NAcc neuron is outlined thus developing an overall framework that has extracellular as well as intracellular components making it biologically realistic. A particular systems physiology study that the author participated in is also reported. Importantly, this thesis lays the groundwork for a systematic study of the reward mechanism from a computational modeling perspective
